چکیده

The mutant strain of Aspergillus niger NG-110 was evaluated for the present investigation of citric acid production. The cultural conditions optimized during the course of study were pH and K4Fe(CN)6, K2HPO4 and CaCl2 concentrations. Cane-molasses was utilized as the basal fermentation medium. The mutant NG-110 produces maximal amount of citric acid when the pH of the medium was 6. When the concentrations of K4Fe(CN)6, K2HPO4 and CaCl2 were 200 ppm, 0.2% w/v and 2.0%, the maximum citric acid production was 69.30±0.8 g/l, 81.21±0.2 g/l and 80.76±0.2 g/l, respectively.

Changes in transcript levels of starch hydrolysis genes and raising citric acid production via carbon ion irradiation mutagenesis of Aspergillus niger

The filamentous ascomycete Aspergillus niger is well known for its ability to accumulate citric acid for the hydrolysis of starchy materials. To improve citric acid productivity, heavy ion beam mutagenesis was utilized to produce mutant A.niger strains with enhanced production of citric acid in this work. It was demonstrated that a mutant HW2 with high concentration of citric acid was isolated ...

EFFECT OF LOW pH ON CONTINUOUS CITRIC ACID FERMENTATION BY ASPERGILLUS NIGER

The present study is concerned with the effect of low pH on continuous citric acid fermentation by Aspergillus niger strain SK-17. Microbial cultivations were carried out in a stainless steel stirred bioreactor of 15 L total capacity. The bioconversion of glucose to citric acid at low pH was investigated on cellulose fabric. Sucrose solution (100 g/l) was made to flow through capillaries of a v...

Investigation on Biosolubilization of Coal by Aspergillus Niger

The effect of demineralization on Indian bituminous coal by filamentous fungus Aspergillus niger has been investigated. X-ray diffraction profile reveals the presence of inorganic components in the sample. Biosolubilised products were characterized by FT-IR, SEM-EDS and XRD. The study reveals that coal consist of a semi-crystalline turbostratic structure. The broad diffraction peak at 20~25.5 a...

Optimization of Citric Acid Production Using Aspergillus niger Isolated from the Leaf Litter Soil of Sathuragiri Hills

Soil samples were collected from the different locations in Sathuragiri Hills (Western Ghats, Tamilnadu). The Total Fungal Counts in the soil samples of the different locations were analyzed. The percentage frequencies of occurrence of fungal isolates are identified. The fungal isolates were identified based on the morphological and cultural characteristics they were identified as Cladosporium ...

Inhibition of citric acid accumulation by manganese ions in Aspergillus niger mutants with reduced citrate control of phosphofructokinase.

Mutant strains of Aspergillus niger with reduced citrate control of carbohydrate catabolism (cic mutants) grow faster than the parent strain on media containing 5% (wt/vol) citrate. The mutants tolerated a higher intracellular citrate concentration than the parent strain. One mutant (cic-7/3) contained phosphofructokinase activity significantly less sensitive towards citrate than the enzyme fro...
